In Belgium, Tanakan® is a registered drug used as symptomatic treatment of brain function disorders (such us memory and concentration troubles) occurring during senescence or Alzheimer's disease. Its active compound corresponds to Ginkgo biloba (L.) dry extract - also called «EGb 761" - obtained from the leaves of the tree. The efficacy of Ginkgo biloba (L.) has not yet been clearly elucidated: a lot of clinical trials have already been published hut they didn’t give the same results. New assessments should be undertaken to achieve solid conclusions.This work describes the manufacturing processes allowing the production of adequate Tanakan, tablets (analysis of raw materials “in-process ·· controls...).It also contains analytical methods which ensure the quality of finished product. Randomized samples during the production process showing conform results according to specifications will enable the release and the marketing of the drug.The methodology of each test is usually described in the European Pharmacopoeia (tablet disintegration, microbiological quality...;. Identification and assays procedures of the active ingredient have to be adjusted specifically to Tanakan because a dedicated analysis method of this drug has never been published. The dosage of active compounds (flavonoids, ginkgolides, bilobalide and ginkgolic acids [limited content] is performed by high performance liquid chromatography with UV or IR detection. The packaging system has been studied and details about product stability and analytical methods validation were discussed. Le Tanakan® est un médicament enregistré en Belgique dans le traitement symptomatique des troubles des fonctions cérébrales (mémoire et concentration) survenant au cours de la sénescence ou de la maladie d'Alzheimer. Son principe actif est l'extrait sec de Ginkgo biloba (Linné) [EGb 761] obtenu à partir des feuilles de l'arbre. L'efficacité du Ginkgo biloba (L) n'a pas encore clairement été démontrée : des dizaines d'études cliniques ont déjà été publiées mais toutes n'arrivent pas aux mêmes conclusions. De nouvelles évaluations devraient être entreprises afin d'aboutir à des résultats solides.Ce mémoire décrit les processus qui permettent une fabrication correcte du Tanakan® (analyse Je matières premières, contrôles en cours de fabrication....). li contient également des méthodes d'analyse afin d'assurer le contrôle-qualité du produit fini. C'est la conformité des résul.tat obtenus à partir d'un échantillon prélevé pendant la production avec les spécifications prévues qui permettra la libération et la commercialisation du lot concerné.Les principes de chaque test sont habituellement décrits dans la Pharmacopée Eurvp6enne (désagrégation des comprimés, qualité microbiologique, ...). En ce qui concerne l'identification et le dosage du principe actif, ils doivent être adaptés spécifiquement au Tanakan® car aucune méthode propre à ce médicament n'a été publiée. L'identification est réalisée pur chromatographie sur couche mince, sur base de standards de référence. Le dosage en principe actif:, (flavonoïdes, ginkgolides, bilobalide et acides ginkgoliques [teneur limite]) est effectué par chromatographie liquide à haute performance avec détection UV ou RI. Le système de conditionnement a été étudié et des informations quant à la stabilité du produit et à la validation des méthodes analytiques employées ont été discutées.

[Voorwoord]Hoewel er sinds 2000 een Europese regelgeving in voege is betreffende de weesgeneesmiddelen zijn zeldzame ziekten, die zoals Mucoviscidose, de ziekte van Gaucher en ongeveer 7000 andere ziekten meestal ernstig en soms levensbedreigend zijn, nog steeds eerder onbekend in België, en onbekend maakt onbemind. Voor patiënten voor wie eindelijk een behandeling beschikbaar is, blijft toegang ertoe een moeilijke en soms dure onderneming.Het symposium wil de huidige situatie betreffende zeldzame ziekten en weesgeneesmiddelen in België overlopen en voorstellen formuleren voor een patiëntgerichte en efficiënte aanpak van deze ziekten, zowel op diagnose als op behandelingsgebied. De verwezenlijkingen in Nederland en Frankrijk staan hiervoor model. Het symposium is bedoeld voor al wie in deze problematiek geïnteresseerd of betrokken partij is, met inbegrip van patiënten, verzorgers, onderzoekers, bio-farmaceutische bedrijven, overheidsinstanties, politici en anderen.[Préface]Bien qu'une régulation Européenne soit en vigueur depuis l'an 2000, les maladies rares, telles que la Mucoviscidose, la maladie de Gaucher et environ 7000 autres maladies, sont peu connues et ne bénéficient pas d'une approche structurée en Belgique. Même pour les patients qui pourraient enfin bénéficier d'un traitement efficace, l'accès à ces traitements reste difficile et souvent coûteux.Le but du symposium du 7 décembre est d'évaluer la situation des maladies rares et des médicaments orphelins en Belgique et de la comparer à celle des Pays-Bas et de la France, qui ont réussi à mettre en place une approche globale. Ce symposium s'adresse à tous ceux qui s'intéressent à cette problématique : patients, soignants, chercheurs, industrie bio-pharmaceutique, instances gouvernementales, responsables politiques et autres.
